Thiamine triphosphoric acid; its relation to the thiamine pyrophosphate (cocarboxylase) of Lohmann and Schuster.

چکیده
By analogy with the designation ATP for adenosine triphosphoric acid, TTP is proposed for the new triphosphoric acid derivative of thiamine. The most significant physiological properties of TTP are concerned with the heart (16, 17, 21), TTP being much more active than thiamine or cocarboxylase. Investigations in the enzymatic field have now been made in order to compare TTP with cocarboxylase and to determine the activity of potato apyrase and adenosine triphosphatase (ATPase) towards TTP.

The Relation of Thiamine to Citric Acid Metabolism*
The discovery of the identity of thiamine pyrophosphate and cocarboxylase and the recognition of the ester as the predominant form of thiamine in yeast (1, 2) and animal tissues (3, 4) has been followed by several papers which have clearly demonstrated that cocarboxylase is essential for the oxidative metabolism of pyruvic acid (5-7). However, it has become clear that pyruvic acid is one of the...
متن کاملThiamine and Cocarboxylase Concentration
In patients with congestive heart failure, thiamine deficiency has been demonstrated by the test-dose method (1, 2), by measurement of the lactate-pyruvate ratio of the blood (3), and by determination of blood cocarboxylase (4). The oxidation of pyruvate to acetate requires thiamine in the form of cocarboxylase (thiamine pyrophosphate). Olson, Pearson, Miller, and Stare have shown that thiamine...
متن کاملAn enzymatic study of the mode of action of pyrithiamine (neopyrithiamine).
Because pyrithiamine (neopyrithiamine)’ calls forth in higher animals many of the characteristic signs of thiamine deficiency (2), and because these manifestations may be prevented or corrected in a competitive fashion by the vitamin, the belief has arisen that the pharmacological properties of this analogue of the vitamin are due to its ability to interfere with the metabolic utilization of th...
متن کاملA Kinetic and Mechanistic Study of Hydrolysis of Thiamine Pyrophosphate (Cocarboxylase) in Aqueous Buffer and Micellar Media
Hydrolysis of Thiamine Pyrophosphate (TPP) is too sluggish even at elevated temperatures in aqueous buffer media. However, the hydrolysis reactions of thiamine pyrophosphate (TPP) are dramatically accelerated under micellar conditions even at room temperature by the addition of anionic (SDS) and nonionic (Tx) micelles. Menger – Portnoy’s enzymatic model and Piszkiewicz co-operativity model were...
